The Blues suffered an away reverse in midweek after an opening day win in the BGB One Midlands Division.
The new-look Blues were beaten 3-1 at Chesham United on Tuesday night.
This was despite taking a first-half lead through Jamie Osborne.
But the hosts levelled before the break and went on to claim all three points.
Arlesey had marked their sideways move from Ryman League Division One North with a comfortable 3-0 home win over Rothwell Town on Saturday.
Matthew Kwomini put them in front after six minutes and the visitors then lost Robbie Maddox who was sent off for elbowing in the 28th minute.
Goals from Ryan Francis (eight minutes after the break) and Kwomini (10 minutes from time) secured the win after the break.
